Target Information

Smoothened drosophila homolog (SMOH) is a serpentine protein belonging to the G-protein coupled receptor Fz/Smo family with 787 amino acids. It is a seven-transmembrane G protein-coupled receptor with four glycosylation sites and a putative extracellular amino terminus. Smoothened, activated by β-Arrestin 2 and GRK2, acts as a receptor for Sonic hedgehog (Shh) and is a positive regulator of the hedgehog signaling pathway along with the patched protein (PTCH). It regulates vertebrate developmental processes, cellular proliferation, and differentiation by activating transcription factor Gli. Mutations in Smoothened are associated with sporadic basal cell carcinoma of the skin. It is widely expressed in heart and brain tissues.
