Target Information

Sterol regulatory element-binding proteins (SREBPs) are transcription factors belonging to the basic helix-loop-helix leucine zipper family of DNA-binding proteins. Three isoforms (SREBP-1a, SREBP-1c, SREBP-2) exist in mammals, differing in structure, regulation, and function.
SREBP-1a and SREBP-1c are products of alternative promoter usage from the SREBP-1 gene, while SREBP-2 originates from a separate gene.
SREBPs are synthesized as 120 kDa inactive precursors in the endoplasmic reticulum membrane. Upon activation, they are translocated to the Golgi, where proteolytic cleavage produces a 60–78 kDa transcriptionally active fragment.
In liver and adipose tissue, SREBPs regulate lipid and cholesterol metabolism by inducing transcription of related genes. SREBP-1 primarily regulates triglyceride synthesis, while SREBP-2 is more closely associated with cholesterol synthesis.
