Target Information
F-Box and WD Repeat Domain Containing 7 (FBXW7) is a member of the F-box protein family characterized by an approximately 40 amino acid motif, the F-box. These proteins form one of the four subunits of the ubiquitin protein ligase complex (SCF: SKP1–Cullin–F-box), functioning in phosphorylation-dependent ubiquitination.
F-box proteins are divided into three classes:
- Fbws: Contain WD-40 domains
- Fbls: Contain leucine-rich repeats
- Fbxs: Contain other interaction modules or no recognizable motifs
FBXW7 (previously FBX30) belongs to the Fbws class and contains seven tandem WD40 repeats. It binds directly to cyclin E and likely targets it for ubiquitin-mediated degradation. Mutations in this gene have been detected in ovarian and breast cancer cell lines, suggesting a role in cancer pathogenesis. Multiple transcript variants encoding different isoforms have been identified.
