Target Information

COMMD1 is a proposed scaffold protein involved in diverse physiological processes, including regulation of ubiquitination of specific cellular proteins. It modulates cullin-RING E3 ubiquitin ligase (CRL) complexes by displacing CAND1, promoting CRL E3 activity and dissociating CAND1 from CUL1 and CUL2.
It promotes ubiquitination and proteasomal degradation of NF-κB subunit RELA, down-regulating NF-κB activity. COMMD1 also regulates membrane expression and ubiquitination of SLC12A2, modulates sodium transport in epithelial cells via ENaC subunits, and promotes CFTR surface expression.
It plays roles in copper ion homeostasis and copper-dependent ATP7A trafficking, interacting with CCC and WASH complexes. COMMD1 binds one copper ion per monomer and phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2), and is involved in HIF1A-mediated transcription regulation.
