Target Information
HEF-1 (also known as NEDD9) is a multifunctional protein involved in integrin-based signaling that regulates cell motility, growth, apoptosis, and oncogenic transformation.
Members of the Cas family, including HEF1, are key substrates of focal adhesion kinase (FAK) and Src family tyrosine kinases—important drug development targets.
Overexpression of HEF1 increases mRNA levels of genes associated with motility and invasiveness (e.g., metalloproteinases, MLCK, p160ROCK, ErbB1) and can induce apoptosis in epithelial-derived cell lines such as MCF7 and HeLa.
Clinical studies indicate that overexpression of the related protein BCAR1 (p130Cas) correlates with tamoxifen resistance, underscoring the importance of this protein family in cancer prognosis.
