Target Information
Xeroderma pigmentosum type G (XPG)는 햇빛에 대한 극도의 민감성을 보이는 인간 유전 질환입니다. XPG 단백질은 flap endonuclease 1 (FEN-1) 구조 특이적 DNA 복구 endonuclease 계열에 속하며, 자외선(UV)에 의해 유도된 DNA 손상을 복구하는 데 필수적인 효소입니다. Human XPG nuclease는 뉴클레오타이드 절제 복구(nucleotide excision repair) 과정에서 3' 절단을 수행합니다. 이 효소는 이중가닥 DNA와 비짝합 DNA의 접합 부위 근처에서 DNA 버블 구조를 절단합니다. Human XPG의 29 아미노산 영역(981–1009)은 PCNA 결합 활성을 가지며, Arg992 잔기가 PCNA 결합에 중요합니다. 또한 Replication Protein A (RPA)는 XPA 및 XPG 단백질과 직접 결합하여 손상 인식 및 절제 복구 과정에 관여합니다.
