Target Information
ACVR2A와 ACVR2B는 세포막에 존재하는 트랜스멤브레인 세린/트레오닌 키나아제로, activin type-1 수용체(ACVR1, ACVR1B, ACVR1C 등)와 복합체를 형성하여 activin 신호를 세포 내로 전달합니다. 이 경로는 신경 분화 및 생존, 모낭 발달, FSH 생성, 상처 치유, 세포외 기질 생성, 면역억제 및 발암 등 다양한 생리적·병리적 과정에 관여합니다.
Activin은 난소의 여포 발달에서 자가 또는 측분비적 역할을 수행하며, type-2 수용체는 activin-A/INHBA, activin-B/INHBB 및 inhibin-A 복합체(INHA-INHBA)를 결합합니다. 활성화된 type-2 수용체는 type-1 수용체를 인산화하여 SMAD2 및 SMAD3 단백질을 활성화시키며, 이들은 SMAD4와 복합체를 형성해 핵으로 이동하여 유전자 발현을 조절합니다.
SMAD7은 ACVR1B에 결합하여 SMAD2/3의 결합을 억제함으로써 activin 신호를 차단할 수 있습니다. 또한 inhibin-B가 IGSF1 coreceptor를 통해 수용체에 결합함으로써 activin 신호전달을 억제하기도 합니다.
