Target Information

COX2 converts arachidonate to prostaglandin H2 (PGH2), a committed step in prostanoid synthesis, including production of inflammatory prostaglandins.
The conversion of arachidonate to PGH2 involves two reactions:

  1. Cyclooxygenase (COX) reaction: converts arachidonate to prostaglandin G2 (PGG2)
  2. Peroxidase reaction: reduces PGG2 to prostaglandin H2 (PGH2)

COX2 is constitutively expressed in some tissues under physiological conditions (endothelium, kidney, brain) and up-regulated under pathological conditions such as cancer and inflammation.
Up-regulation is associated with increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis. In cancer cells, COX2 plays a key role in prostaglandin E2 (PGE2) production, modulating motility, proliferation, and apoptosis resistance.

COX2 is naturally inhibited by calcitriol (active Vitamin D). Glucocorticoids trans-repress PTGS2 gene activity in vivo by interfering with transcription initiation and elongation. COX2 is a target of NSAIDs such as aspirin, which reduce pain and swelling from COX2-driven inflammation.
