Target Information
ATG3 (APG3) is a widely expressed conjugating enzyme essential for APG8 lipidation, a critical step in autophagy initiation. Autophagy enables degradation of cellular proteins via the autophagosomal-lysosomal pathway, supporting normal growth control and nutrient preservation under starvation. Dysregulation of this process may occur in tumor cells. ATG3 acts as an E2-like enzyme forming the Atg8-phosphatidylethanolamine (Atg8-PE) conjugate, crucial for autophagosome formation. The process is negatively regulated by TOR through phosphorylation of autophagy protein APG1.
