Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, playing key roles in regulating cell death and stress responses. TRAF proteins share homology regions enabling receptor and inter-TRAF interactions, activating diverse signaling cascades. For example, TRAF2 and TRAF3 bind CD40 to mediate NF-κB activation in B lymphocytes, while TRAF2 and TRAF6 are required for CD40 signaling in non-hematopoietic cells. TRAF2 also associates with LTβR, TRAF3, and apoptosis inhibitors cIAP1 and Smac.
