Target Information
Cyclin D1 (PRAD-1, bcl-1)는 세포 주기 조절의 핵심 단백질로, cdk4 및/또는 cdk6과 함께 Rb 단백질을 인산화하여 작용합니다. Cyclin D1은 다양한 인간 종양(예: mantle cell lymphoma)에서 과발현되는 원암유전자(proto-oncogene)로 알려져 있습니다.
이 단백질은 G1 phase 동안 합성되며, 성장 인자 자극에 따라 CDK4 또는 CDK6과 복합체를 형성하여 세포 주기의 G1/S 전이를 조절합니다. Cyclin D1은 Rb 단백질과 상호작용하며, p21Cip1 및 p27Kip1과 같은 CDK 억제제의 기능을 조절합니다.
Cyclin D1의 불안정성은 유비퀴틴 매개 분해를 통해 조절되며, 이는 G1/S phase 정지에 관여합니다. Cyclin D1은 세포주기 동안 단백질 양의 주기적 변화를 보이는 highly conserved cyclin family에 속합니다.
