Target Information

MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases and contains an N-terminal catalytic domain and a C-terminal regulatory domain. MAP4K3 acts upstream of the MEKK1 target and is stimulated by UV radiation and TNF-alpha, leading to activation of the JNK signaling pathway. It can directly activate PKC-θ during TCR signaling, and elevated MAP4K3 expression is observed in T cells from patients with systemic lupus erythematosus. MAP4K3 functions as a pro-apoptotic kinase that activates BAX through posttranscriptional modulation of PUMA, BAD, and BIM. The encoded protein’s kinase activity is stimulated by UV radiation and TNF-alpha and specifically activates the c-Jun N-terminal kinase (JNK) pathway. It functions upstream of mitogen-activated protein kinase kinase 1 (MEKK1). Previously known as RAB8-interacting protein-like 1 (RAB8IPL1), MAP4K3 has been renamed mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
