Target Information

TLR4 is a member of the Toll-like receptor (TLR) family, which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. TLR4 is most abundantly expressed in the placenta and the myelomonocytic subpopulation of leukocytes. Mammalian cells respond to LPS by activating TLR4, which forms part of a multi-protein complex containing CD14, LY96, and TLR4, involved in signal transduction induced by lipopolysaccharides from Gram-negative bacteria.

TLR4 recognizes pathogen-associated molecular patterns (PAMPs) and mediates cytokine production necessary for effective immunity. Mutations in the TLR4 gene are associated with differences in LPS responsiveness. Several transcript variants of the TLR4 gene exist, though most have uncertain protein-coding potential. TLR4 is expressed by peripheral blood monocytes, a small population of B-cells, and in human placenta. Studies using TLR4-deficient mice show increased susceptibility to Gram-negative sepsis, confirming its key role in LPS recognition.
