Target Information

Complex V, also known as F1F0 ATPase or ATP synthase, is responsible for ATP production in oxidative phosphorylation and can also function in reverse as a proton pumping ATPase.
Originally thought to be localized exclusively to mitochondria, it has also been identified on the plasma membrane of several cell types:

  • On hepatocytes, it functions as the HDL receptor.
  • On endothelial cells, it may act as the angiostatin receptor.
  • On the surface of cancer cells, it has additional functional roles.

In mammals, the enzyme is composed of 17 subunits:

  • Five subunits form the detachable F1 portion.
  • The remaining subunits are part of two stalk domains and the proton-pumping F0 portion.
  • Two F0 subunits are encoded on mitochondrial DNA, while the others are nuclear encoded.

Mutations in mitochondrial-encoded subunits of ATP synthase (Complex V) are associated with OXPHOS disease.
