The Oncomine™ TCR Beta-SR Assay - DNA is a robust, targeted next-generation sequencing (NGS) assay designed to accurately identify and measure the clonal expansion of T lymphocytes in formalin-fixed paraffin-embedded (FFPE) tumor samples.
This assay identifies unique T-cell clones by targeting the diverse complementarity-determining region 3 (CDR3) of the T-cell receptor (TCR) gene locus in genomic DNA. The nucleotide sequence of the CDR3 region is unique to each T-cell clone and codes for the part of the TCR beta chain involved in antigen recognition.

워크플로우 및 응용 분야

The assay uses multiplex PCR primers to generate 80-bp amplicons that can be sequenced on all chip types supported by the Ion GeneStudio™ S5 sequencing systems.
The workflow—from DNA isolation to data analysis—can be completed within two days using the Ion Chef™ templating system and Ion GeneStudio™ S5 system.

This assay supports key applications in immuno-oncology and hemato-oncology research.
Its high sensitivity, flexible input requirements, and high-throughput capability make it ideal for minimal residual disease testing in T-cell malignancies.
The FFPE compatibility enables analysis of T-cell clonality in the tumor microenvironment, useful for biomarker discovery and monitoring responses to immunotherapies such as checkpoint inhibitors, cancer vaccines, and CAR-T cell therapies.
