Target Information

Various biochemical, physiological, and behavioral processes display circadian rhythms controlled by an internal biological clock. The central mechanisms driving this clock are composed of an autoregulatory transcription/post-translation-based feedback loop. Cryptochrome 1 (CRY1) and 2 (CRY2) are DNA-binding flavoproteins homologous to blue-light receptors and photolyases.
In Drosophila, CRY acts as a photoreceptor for the circadian clock, binding to TIM in a light-dependent manner to regulate its function. Mammalian CRY1 and CRY2 interact with circadian genes CLOCK and BMAL1, as well as PER1, PER2, and TIM, functioning as light-independent components of the circadian clock. They regulate Per1 transcriptional cycling through interactions with activators and inhibitors.
Mutant mice lacking Cry1 or Cry2 show altered locomotor periodicity, indicating that both proteins are essential for synchronizing circadian phases. Double-mutant mice exhibit arrhythmicity in darkness, demonstrating the necessity of Cry1 and Cry2 balance for proper clock function.
