Target Information

DNA methylation, or the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and gene expression regulation. The methylation pattern involves enzymatic methylation and demethylation processes.
The mammalian demethylation enzyme exhibits activity associated with a methyl-CpG-binding domain (MBD), catalyzing the removal of methyl groups as methanol. MeCP2 and MBD1 repress transcription by binding to methylated DNA, while MBD2 and MBD4 (MED1) colocalize with heavily methylated satellite DNA, mediating methylation signal functions.
MBD3 does not bind methylated DNA in vivo or in vitro. MBD1–MBD4 are expressed in somatic tissues, but MBD1 and MBD2 expression is reduced in embryonic stem cells, which lack MeCP1 activity. MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and is mutated in some microsatellite unstable tumors.
