Target Information
Ceramides are synthesized during epidermal differentiation and accumulate within the stratum corneum, forming key components of the epidermal permeability barrier. Excess ceramide can induce antimitogenic signaling and apoptosis, while ceramide metabolites such as sphingosine and sphingosine-1-phosphate (S1P) play important bioregulatory roles.
Ceramide hydrolysis in nucleated cell layers regulates keratinocyte proliferation and apoptosis under external stress. At the stratum corneum, ceramide hydrolysis releases free sphingoid bases that act as endogenous antimicrobial agents.
ACER1 is highly expressed in the epidermis and catalyzes hydrolysis of very long-chain ceramides to generate sphingosine.
