Target Information

Influenza A H1N1 NS1 is a non-structural protein that inhibits post-transcriptional processing of cellular pre-mRNA by binding and inhibiting two cellular proteins required for 3'-end processing: CPSF4 and PABPN1.
This leads to accumulation of unprocessed pre-mRNAs that cannot be exported from the nucleus, resulting in early shutdown of cellular protein synthesis after infection.
Viral protein synthesis remains unaffected as viral mRNAs are polyadenylated by the viral polymerase through a stuttering mechanism.
NS1 also inhibits TRIM25-mediated DDX58 ubiquitination, preventing activation of type I IFN genes.
Additionally, NS1 binds poly(A) and U6 snRNA and suppresses RNA silencing-based antiviral responses in Drosophila cells.
It may also prevent EIF2AK2/PKR activation by binding double-strand RNA or directly interacting with EIF2AK2/PKR, a mechanism relevant early in infection when NS1 is mainly cytoplasmic.
