Target Information
The COL1A1 gene encodes the pro-alpha1 chain of type I collagen, which forms a triple helix comprising two alpha1 chains and one alpha2 chain. Type I collagen is a fibril-forming collagen found in most connective tissues and is abundant in bone, cornea, dermis, and tendon.
Mutations in COL1A1 are associated with:
- Osteogenesis imperfecta types I–IV
- Ehlers-Danlos syndrome type VIIA and Classical type
- Caffey Disease
- Idiopathic osteoporosis
Reciprocal translocations between chromosomes 17 and 22 (COL1A1 and PDGFB genes) are linked to dermatofibrosarcoma protuberans, causing unregulated growth factor expression. Two transcript variants have been identified using alternate polyadenylation signals.
