Product Specific Information
- Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 minutes.
Target Information
ZAP-70 is a cytosolic protein tyrosine kinase belonging to the Syk family. It is expressed exclusively in T-cells and natural killer cells and is essential for T-cell receptor activation.
Upon T cell antigen receptor (TCR) engagement, ZAP70 is phosphorylated on tyrosines 292, 315, and 319 in the interdomain B region between the SH2 and kinase domains.
Phosphorylation of tyrosines 315 (Vav binding site) and 319 (Lck binding site) enhances ZAP70-mediated lymphocyte signaling, while tyrosine 292 phosphorylation attenuates signaling.
