• For Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and facilitating iron recycling.
CD163 is expressed on human monocytes and macrophages, particularly during late inflammation phases. It is present on all circulating monocytes and most tissue macrophages except those in mantle zones, germinal centers, interdigitating reticulum cells, and Langerhans cells.
Expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding of CD163 into plasma. CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner and triggers intracellular signaling cascades involving calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
