Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA), expressed on all hematopoietic cells except mature erythrocytes and platelets. CD45 functions as a tyrosine phosphatase and is key in lymphocyte activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse bone marrow cells at ≤0.4 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD45 (Leukocyte Common Antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent from non-hematopoietic cells and tissues. CD45 regulates Src-family kinases, T- and B-cell receptor signaling, cytokine receptor signaling, and integrin-mediated pathways. Isoforms (e.g., CD45RA) arise from alternative splicing and vary by cell type and physiological state.
