Background
The androgen receptor gene spans more than 90 kb and encodes a protein with three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. The receptor acts as a steroid-hormone activated transcription factor. Upon ligand binding, it dissociates from accessory proteins, translocates to the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
This gene includes two polymorphic trinucleotide repeat segments encoding polyglutamine and polyglycine tracts. Expansion of the polyglutamine tract from normal 9–34 repeats to 38–62 repeats leads to spinal bulbar muscular atrophy (Kennedy disease). Mutations in this gene are also associated with complete androgen insensitivity (CAIS). Two alternatively spliced variants encode distinct isoforms.
