세포 내 위치 (Cellular Localization)
Secreted; extracellular space and extracellular matrix. In plasma, APOE associates with chylomicrons, remnants, VLDL, LDL, and HDL lipoproteins (PubMed:1911868, 8340399). Lipid-poor oligomeric APOE binds extracellular matrix in a calcium- and heparan-sulfate proteoglycan-dependent manner (PubMed:9488694). Lipidation induces release from the matrix.
배경 (Background)
The APOE gene encodes a major apoprotein of chylomicrons. It binds to liver and peripheral cell receptors and is essential for normal catabolism of triglyceride-rich lipoproteins. The gene is located on chromosome 19 alongside apolipoprotein C1 and C2 genes. Mutations cause familial dysbetalipoproteinemia (Type III hyperlipoproteinemia), resulting in elevated plasma cholesterol and triglycerides due to impaired clearance of chylomicron and VLDL remnants.
