Background
- Developmental stage: Expressed in early stages of myeloid and erythroid differentiation.
- Function: Substrate of antigen receptor-coupled tyrosine kinase; involved in antigen receptor signaling for clonal expansion and deletion in lymphoid cells; may regulate gene expression.
- Post-translational modification: Phosphorylated by LYN rapidly after cross-linking of surface IgM on B-cells.
- Similarity: Contains one SH3 domain and four cortactin repeats.
- Subunit: Associates with SH2 and SH3 domains of LCK. Binding to LCK SH3 domain occurs constitutively, while SH2 binding occurs upon TCR stimulation. Similar binding observed with LYN, but not with FYN. Directly associates with HAX1 through its C-terminal region and interacts with HS1BP3.
- Tissue specificity: Expressed only in tissues and cells of hematopoietic origin.
