Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and has been identified as a co-receptor for HIV-2 binding via gp120, promoting virus-cell fusion. CXCR4 also serves as the receptor for C-X-C chemokine CXCL12/SDF-1, mediating intracellular calcium signaling and MAPK1/MAPK3 activation.

Additionally, CXCR4 acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP levels. It plays critical roles in hematopoiesis, cardiac development, and vascularization of the gastrointestinal tract. In the CNS, CXCR4 contributes to hippocampal neuron survival and cerebellar development. It binds bacterial lipopolysaccharide (LPS) and mediates LPS-induced inflammatory responses, including TNF secretion by monocytes. Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection of human cells. The N-terminal domain and second extracellular loop of CXCR4 serve as HIV binding sites.
