Target Information
HERC5 (E3 ISG15-protein ligase HERC5) is a major E3 ligase for ISG15 conjugation and acts as a positive regulator of the innate antiviral response induced by interferon. It functions as part of the ISGylation machinery that recognizes target proteins broadly and catalyzes ISGylation of IRF3, resulting in sustained activation. HERC5 attenuates IRF3-PIN1 interaction, antagonizes IRF3 ubiquitination and degradation, and enhances antiviral response. It is associated with polyribosomes and modifies newly synthesized proteins in a cotranslational manner.
