Target Information
Calcium/calmodulin-dependent protein kinase that functions autonomously after Ca(2+)/calmodulin-binding and autophosphorylation. It is involved in dendritic spine and synapse formation, neuronal plasticity, and regulation of sarcoplasmic reticulum Ca(2+) transport in skeletal muscle. In neurons, it plays a structural role in actin cytoskeleton reorganization during plasticity by binding and bundling actin filaments in a kinase-independent manner. This function is critical for proper targeting of CaMK2A, which promotes dendritic spine and synapse formation and maintains synaptic plasticity enabling long-term potentiation (LTP) and hippocampus-dependent learning. In developing hippocampal neurons, it promotes dendritic arborization, and in mature neurons, dendritic remodeling. It also modulates skeletal muscle function in response to exercise, regulating SR Ca(2+) transport in slow-twitch muscles and controlling Ca(2+) release in fast-twitch muscles through phosphorylation of triadin and phospholamban.
