Invitrogen™ Acetylcholinesterase Fluorescent Activity Kit
The Acetylcholinesterase Fluorescent Activity Kit is a research-use-only fluorescent assay designed for the quantification and detection of acetylcholinesterase (AChE) activity in serum, plasma, and erythrocyte membrane samples.
This complete, ready-to-use kit includes:

Background
Acetylcholine (ACh) is an essential neurotransmitter in both the central and peripheral nervous systems. It mediates chemical neurotransmission through nicotinic and muscarinic receptors found in neurons, ganglia, heart, smooth muscle fibers, and glands.
Acetylcholinesterase (AChE), encoded by the single AChE gene, exhibits structural diversity due to alternative mRNA splicing and post-translational modifications.
- The hydrophilic form predominates in brain and muscle tissues, forming disulfide-linked oligomers with structural subunits.
- The erythroid form contains a cleavable peptide with a GPI anchor, associating with membranes via phosphoinositide moieties.
This assay is validated for serum, EDTA and heparin plasma, and solubilized RBC ghosts from multiple species.
Assay Principle
The kit quantitatively measures AChE activity using a proprietary non-fluorescent molecule that binds covalently to the thiol product of the enzymatic reaction between AChE substrate and enzyme.
This reaction yields a fluorescent product detectable at 510 nm (excitation 390 nm) using a fluorescence plate reader.
A human AChE standard is included to generate a standard curve for accurate quantification.
The assay is suitable for:
- Diluted serum and plasma
- RBC ghosts
- Extracted tissue samples
- Cell lysates
AChE Unit Definition
One unit hydrolyzes 1.0 μmol of acetylthiocholine iodide per minute at pH 7.4 and 37°C.
