Target Information
Spleen tyrosine kinase (SYK) is a cytoplasmic tyrosine kinase, along with ZAP70, involved in immune receptor signaling. SYK translocates to the plasma membrane upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, phosphorylating downstream adaptor proteins to initiate signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription. SYK binds receptor assemblies via SH2 domain interactions with phosphorylated ITAM motifs, and is activated by Src-family kinases.
