Target Information
Human immunodeficiency virus (HIV) and related viruses require coreceptors, in addition to CD4, to infect target cells. CCR5, CXCR4, CCR3, CCR2b, and CCR8 are G protein-coupled receptors that serve as HIV coreceptors.
CCR5 (CC-CKR-5) acts as a principal coreceptor for macrophage- and dual-tropic HIV-1 strains, mediating fusion and entry into human white blood cells. It is essential for infection by HIV-1, HIV-2, and SIV.
The beta-chemokines RANTES, MIP-α, and MIP-β are natural ligands for CCR5 and can prevent infection by M-tropic HIV-1. CCR5 interacts with the CD4–gp120 complex on HIV, facilitating membrane fusion and viral entry.
CCR5 mRNA is expressed in lymphoid organs and monocytes, with the amino-terminal domain and extracellular loops serving as HIV binding sites.
