Target Information
Ago proteins are broadly expressed in somatic cells, associate with miRNAs, and are key actors in RNA silencing pathways. Only Ago2 displays endonucleolytic or “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when perfect base-pairing occurs. With partial complementarity, Ago2 instead represses translation. Ago2 plays a non-redundant role in small RNA-guided gene silencing processes, including RNA interference, translation repression, and heterochromatinization. As a core element of the RISC complex, AGO2 initiates degradation of target mRNAs through catalytic activity guided by siRNAs or miRNAs. Additionally, Ago2 acts as an RNA slicer in a Dicer-independent manner and regulates miRNA maturation. Overexpression of Ago2 correlates with cancer progression and patient survival. Gene disruption studies in mice show Ago2 is essential for embryonic development and regulates B-lymphoid and erythroid development.
