Target Information
CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. Originally identified on Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1, CD30 interacts with its ligand CD30L (CD153) to mediate effects such as cell proliferation, activation, differentiation, and apoptosis.
CD30 plays a crucial role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas, acting as a costimulatory molecule in thymic negative selection. It is expressed on Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (including Burkitt’s), virus-infected T/B cells, and activated normal T/B cells.
In T cells, CD30 is found on subsets producing Th2-type cytokines and on CD4⁺/CD8⁺ thymocytes co-expressing CD45RO and IL4 receptor. Soluble CD30 (sCD30) serves as a marker of Th2 immune response. TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling, regulating apoptosis and limiting autoreactive CD8 effector T cell proliferation. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
