MLCK (MLCK2), also known as MYLK3, belongs to the myosin regulatory light chain kinases that facilitate muscle contraction. As a member of the Ser/Thr protein kinase family, MLCK phosphorylates a specific serine in the N-terminus of myosin light chains (MLC), enabling interaction with actin filaments. This process is crucial for smooth muscle contraction and plays a central role in vascular permeability and tissue edema formation.

In the nervous system, MLCK regulates astrocytic process growth and participates in transmitter release at sympathetic ganglion synapses. It also contributes to fibroblast apoptosis signaling. Smooth muscle and non-muscle isozymes are expressed in diverse adult and fetal tissues, with non-muscle isoform 2 being the dominant splice variant. The Telokin isoform binds calmodulin and is found in various tissues. MLCK activity is likely down-regulated by phosphorylation. The protein includes one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
