Target Information
Monocarboxylates such as lactate and pyruvate play an integral role in cellular metabolism. Lactic acid is produced in large quantities via glycolysis, providing ATP under normal physiological conditions. Accumulation of lactic acid decreases intracellular pH and halts glycolysis; thus, lactic acid must be transported out of the cell.
This process is mediated by the monocarboxylate transporter (MCT) family, functioning as proton symports and stereoselective for L-lactate. The MCT family includes at least eight members (MCT1–8), each containing 10–12 transmembrane-helical domains with cytoplasmic amino and carboxy termini.
Defects in the SLC16A2 gene encoding MCT8 can cause monocarboxylate transporter 8 deficiency (MCT8 deficiency), leading to impaired hormone transport, X-linked psychomotor retardation, and abnormal thyroid levels.
