Target Information
GAD-65와 GAD-67은 group II decarboxylase 계열의 단백질로, L-glutamic acid로부터 GABA(γ-aminobutyric acid)를 생성하는 과정의 속도 제한 단계를 촉매합니다.
두 단백질 모두 뇌에서 발견되지만, GAD-65는 신경 말단의 시냅스 소포막에 국한되어 있으며, GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성을 담당하며, 신경전달 시 GABA 수요가 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 보조합니다.
GAD-65의 손실은 GABA 신경전달에 손상을 초래할 수 있고, GAD-67의 손실은 치명적입니다.
또한 GAD-67은 alternative splicing으로 인해 주요 형태인 GAD-67과 소수 형태인 GAD-25 두 가지 isoform을 가집니다.
