• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

CD56 (NCAM, neural cell adhesion molecule) is a transmembrane glycoprotein of the immunoglobulin family that acts as an adhesive molecule. It is expressed in the nervous system in isoforms ranging from 120–180 kDa and found on T cells and NK cells. CD56 participates in cell migration, axonal growth, pathfinding, and synaptic plasticity. Polysialic modification reduces CD56-mediated cell adhesion. Through its extracellular region, CD56 mediates homophilic and heterophilic interactions by binding extracellular matrix components such as laminin and integrins. It is expressed on most neuroectodermal-derived cell lines, tissues, and neoplasms (retinoblastoma, medulloblastoma, astrocytomas, neuroblastoma). CD56 serves as a sensitive neuroendocrine marker for neuroendocrine and ovarian granulosa cell tumors. Diseases associated with CD56 dysfunction include rabies and blastic plasmacytoid dendritic cell neoplasms.
