Target Information
IKK alpha/beta는 인간의 CHUK (conserved helix-loop-helix ubiquitous kinase) 및 IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) 유전자에 의해 각각 염색체 10번과 8번에 위치하여 발현됩니다.
NF-κB 경로를 활성화하는 TNF-α 등의 자극 시, IKK complex의 두 서브유닛인 IKKα와 IKKβ는 조절 단백질 NEMO (IKKγ)와 함께 IkB 단백질을 인산화하여 유비퀴틴 매개 분해를 유도하고 NF-κB의 핵 내 이동을 촉진합니다.
활성 루프의 Serine 176 및 180의 인산화는 키나아제 활성화를 위해 필수적인 구조 변화를 유도합니다.
