Product Specific Information

This 4D10.1 monoclonal antibody recognizes human Syk, a 72-kDa member of the Syk/ZAP-70 family of non-receptor protein tyrosine kinases. Syk is highly expressed in B cells, with lower expression in immature thymocytes, mast cells, and platelets. It can also be detected in fibroblasts, epithelial cells, hepatocytes, endothelial cells, and neuronal cells.
Syk plays a key role in signaling cascades downstream of B and T cell antigen receptors and is essential for lymphocyte development. Upon recruitment to ITAMs on antigen receptors, Syk is activated by phosphorylation and subsequently phosphorylates proteins such as phospholipase C gamma and BLNK/SLP-65. Abnormal Syk expression has been linked to tumor cell migration and invasion in several cancers.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated using normal human peripheral blood cells with the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Cell number per test may vary from 10^5 to 10^8 cells.

Target Information

Spleen tyrosine kinase (SYK) and ZAP70 are cytoplasmic tyrosine kinases implicated in allergic disorders, autoimmune diseases, and cancer. SYK translocates to the plasma membrane upon BCR or IgE receptor triggering and phosphorylates adaptor proteins, initiating downstream signaling such as calcium mobilization, cytoskeleton remodeling, and gene transcription. SYK interacts with phosphorylated ITAM motifs via its SH2 domains, with Src-family kinases contributing to its activation.
