Target Information
Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) that detects viral double-stranded RNA (dsRNA) and triggers the innate immune response. Along with MDA5, RIG-I functions as an RNA helicase with overlapping and distinct roles.
RIG-I recognizes dsRNAs lacking a 5'-triphosphate end and short dsRNAs, while MDA5 detects long dsRNAs. Upon activation, both signal through IPS-1 to activate transcription factors NF-κB and IRF-3, leading to apoptosis, cytokine signaling, and inflammation.
RIG-I is essential for antiviral responses against influenza A/B, RSV, paramyxoviruses, Japanese encephalitis virus, and West Nile virus. MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
Recent studies also suggest RIG-I involvement in cytosolic DNA detection via RNA polymerase III activity.
