Target Information
The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of two complexes: a 20S core and a 19S regulator.
- The 20S core consists of four rings of 28 non-identical subunits (two rings of 7 alpha subunits and two rings of 7 beta subunits).
- The 19S regulator includes a base (6 ATPase subunits and 2 non-ATPase subunits) and a lid (up to 10 non-ATPase subunits).
Proteasomes are abundant in eukaryotic cells and cleave peptides in an ATP/ubiquitin-dependent, non-lysosomal process. The immunoproteasome variant is essential for MHC class I peptide processing.
The PSMD2 gene encodes one of the non-ATPase subunits of the 19S regulator lid, potentially involved in the TNF signaling pathway via interaction with the TNF type 1 receptor. A pseudogene is located on chromosome 1.
