Target Information

Folylpoly-gamma-glutamate synthetase (FPGS) catalyzes the addition of several equivalents of glutamic acid to the gamma-carboxyl group in the side chain of folate cofactors and analogs.
FPGS plays a key role in folate homeostasis by:

1. Preventing efflux of folate cofactors from the cell through polyglutamation
2. Increasing binding of folate cofactors to enzymes involved in folate interconversion and biosynthesis
3. Allowing accumulation of folates in mitochondria required for glycine synthesis

FPGS is widely expressed in human tumors and is associated with cell proliferation or lack of differentiation.
Its activity enhances the cytotoxic efficiency of folate antimetabolites used in chemotherapy.
