Target Information

MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating the stability of p53. It is located on chromosome 12q. Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, promoting MDM2-mediated ubiquitination and degradation of p53. Phosphorylation also prevents MDM2 binding to p19ARF, further increasing p53 degradation. MDM2 negatively regulates p53 function by inhibiting its transactivation and promoting degradation through its E3 ubiquitin ligase activity. Overexpression of MDM2 leads to p53 inactivation and reduced tumor suppressor function. MDM2 is implicated in apoptosis, cell cycle regulation, and is overexpressed in various malignancies such as soft tissue carcinomas and breast cancer. It interacts with proteins including retinoblastoma 1 and ribosomal protein L5. Over 40 alternatively spliced transcript variants of MDM2 have been identified in both tumor and normal tissues.
