Target Information

The cystatin superfamily is a well-established family of cysteine protease inhibitors.

• Type 1 cystatins (A, B): mainly intracellular
• Type 2 cystatins (C, D, E/M, F, G, S, SN, SA): extracellular
• Type 3 cystatins (kininogens): intravascular proteins

All true cystatins inhibit cysteine peptidases of the papain family, such as cathepsins, and some also inhibit legumain family enzymes.
Cystatin SA, S, and SN are primarily found in saliva, and also expressed in tears, urine, and seminal fluid.
Cystatin C is expressed in brain, thymus, ovary, epididymis, and vas deferens.
Cystatin D protects against oral cavity proteinases, while Cystatin E/M and F modulate cathepsin inhibition.
Fetuins, part of the cystatin superfamily, influence osteogenesis, bone resorption, and regulation of insulin and hepatocyte growth factor receptors.
High and low molecular weight kininogens inhibit thrombin- and plasmin-induced thrombocyte aggregation.
