Cortactin (phospho Tyr421) rabbit pAb

Cytoplasm, cytoskeleton, lamellipodium, ruffle, dendrite, cell membrane (peripheral membrane protein, cytoplasmic side), podosome, cell junction, focal adhesion, clathrin-coated pit, dendritic spine, and cell cortex.
Colocalizes transiently with PTK2/FAK1 at focal adhesions. Associated with membrane ruffles and lamellipodia.
In the presence of CTTNBP2NL, colocalizes with stress fibers. With CTTNBP2, localizes at the cell cortex.
Upon neuronal activation by glutamate, redistributes from dendritic spines to the dendritic shaft. Colocalizes with DNM2 at the basis.

Background

Cortactin (CTTN) is overexpressed in breast cancer and squamous cell carcinomas of the head and neck. The encoded protein localizes in the cytoplasm and at cell-substratum contacts. It plays dual roles in regulating adherens-type junction interactions and organizing the cytoskeleton and cell adhesion structures of epithelial and carcinoma cells. During apoptosis, cortactin is degraded in a caspase-dependent manner. Aberrant regulation contributes to tumor cell invasion and metastasis. Three splice variants encoding different isoforms have been identified (RefSeq, May 2010).
