Background
This gene encodes a member of the doublecortin family. The encoded protein is a cytoplasmic protein containing two doublecortin domains that bind microtubules. In the developing cortex, cortical neurons migrate long distances to reach their differentiation site. The protein directs neuronal migration by regulating microtubule organization and stability. It interacts with LIS1, the regulatory gamma subunit of platelet activating factor acetylhydrolase, essential for proper microtubule function in the developing cortex. Mutations in this gene cause abnormal neuronal migration and cortical layering defects, leading to epilepsy, mental retardation, subcortical band heterotopia (“double cortex” syndrome) in females, and lissencephaly (“smooth brain”).
