| Cellular Localization | Nucleus; Chromosome (centromere, kinetochore); Cytoplasm (spindle pole, centrosome). Localizes to the inner centromere throughout prophase until metaphase and disappears at anaphase. Localization requires BUB1 and PPP2R1A interaction. Colocalizes with NEK2 and SS18L1 at the kinetochore. Phosphorylation by AUKRB and presence of BUB1 are required for kinetochore localization. Isoform 1 primarily localizes to kinetochores during G2 phase and mitosis. |
Background
The protein encoded by this gene belongs to the shugoshin family, which protects centromeric cohesin from cleavage during mitotic prophase by preventing phosphorylation of a cohesin subunit. Reduced expression leads to premature loss of centromeric cohesion, mis-segregation of sister chromatids, and mitotic arrest. It also protects cohesin along chromosome arms during mitotic prophase. An isoform lacking exon 6 is involved in centriole cohesion. Mutations are associated with Chronic Atrial and Intestinal Dysrhythmia (CAID) syndrome, characterized by Sick Sinus Syndrome (SSS) and Chronic Intestinal Pseudo-obstruction (CIPO) (PMID:25282101).
