Description:
The HIT8a monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is found on most thymocytes and subsets of mature T cells and NK cells. It binds MHC class I molecules and, via association with p56lck, contributes to T cell development and activation.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per test in 100 µL final volume. Applicable for 10⁵–10⁸ cells/test.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a key role in signal transduction and T cell activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/β), acting as a co-receptor with TCR for MHC class I molecules. CD8α is essential for MHC-I binding and is also found as homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial in pathogen and tumor cell clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
