EGFRvIII는 EGF 계열 리간드와 결합하는 수용체 티로신 키나제에 포함됩니다. 리간드 결합은 수용체의 이합체화 및 자가 인산화를 유도하여 세포 내 신호 전달 경로를 활성화합니다. 주요 하위 신호 전달 경로에는 RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, STATs 모듈이 포함되며, NF-κB 경로도 활성화할 수 있습니다. 또한 RGS16, MUC1 등의 단백질을 직접 인산화하여 G 단백질 연계 신호전달과 상호작용합니다.
